A 47-marker immune profiling flow cytometry assay to enable comprehensive antigen-specific immune analysis

Abstract The complex nature of the immune system requires deep interrogation at single-cell level. Flow cytometry using CyTOF® technology employs antibodies tagged with isotopically enriched metals instead fluorophores, resulting in discrete signals that facilitate highly multiparametric characterization on a CyTOF enables analysis over 50 parameters simultaneously without impediments spectral overlap or autofluorescence, allowing for reliable evaluation responses. Maxpar® Direct™ Immune Profiling Assay™ is pre-titrated, dried-down, and validated 30-marker antibody cocktail single-tube staining human whole blood PBMC acquisition systems. Using assay Maxpar Pathsetter™ software, stained samples are automatically resolved into 37 populations. In this study, we highlight customizability Direct panel by adding an array 17 surface intracellular T Cell Activation Expansion Panel live-cell barcoding. simultaneous detection key markers expanded enabled comprehensive cell activation antigen-specific recall responses to stimulated pool immunodominant microbial peptides. Taken together, 47-marker automated unlocked deeper immunophenotyping functional profiling activated cells provided insight cell-mediated adaptive response foreign targets.
